Cytopathic assay
CPEs are important aspects of a viral infection in diagnostics. Many CPEs can be seen in unfixed, unstained cells under the low power of an optical microscope, with the condenser down and the iris diaphragm partly closed. However, with some CPEs, namely inclusion bodies, the cells must be fixed and stained then viewed under light microscopy. Some viruses' CPEs are characteristic and therefore can be an important tool for virologists in diagnosing an infected animal or human… WebHere, we describe a cytopathic effect (CPE)-based drug screening assay for SARS-CoV-2 which allows for rapid assessment of drug compound libraries through pre- or posttreatment drug screening procedures and evaluation using a light microscope.

WebLaboratory diagnosis of viral infections can be carried out by many methods. These methods include (a) demonstration of virus-induced cytopathic effects (CPEs) in the cells, (b) direct detection of viruses, (c) virus isolation and viral assays, (d) detection of viral proteins and other enzymes, (e) detection of viral genome, and WebApr 29, 2024 · Assay Overview. WebAt the end of the exposure, the Viral ToxGlo assay was used to measure ATP, which serves as an indicator of cell viability. The TCID 50 or 50% cytopathic effect (CPE) is the virus dilution that reduces the measured cell viability by 50%. This value was used for subsequent studies on the potency of antiviral agents. WebHere, we provide the detailed protocol for the cytopathic effect (CPE) assay and the plaque assay for human coronaviruses 229E (HCoV-229E), HCoV-OC43, and HCoV-NL63, to identify novel antivirals against HCoVs. Neutral red was used in the CPE assay, as it is relatively inexpensive and more sensitive than other reagents.
WebThe Cytopathic Effect (CPE) Assay The CPE assay is recommended by the World Health Organization 38 for the measurement of NAbs and is considered the gold standard method against which all other methods should be compared. 17 In the assay, a virus-susceptible cell line is treated with patient serum and IFNβ prior to treatment with the virus.
